Design Notes

To better make it work in Lactobacillus delbrueckii bulgaricus, we have optimized the codon in our gene circuit. Moreover, to make it easy to insert into the plasmid pET-28a(+), we add restriction enzyme digestion site BamHI ahead and HindIII in the end.However, these two restriction sites were not present in the full sequence design.

This part is found in Escherichia coli. The sequence can be found in the article Programmed cell death in bacteria: proteic plasmid stabilization systems (Ogura & Hiraga, 1983). The sequence can also be found in GenBank. What have to be illustrated is that we have optimized the codon in our gene circuit.
